Several problems needing attention in cell separation:

1) If you want to do cell culture, remember that the reagents (separation solution, washing solution, etc.) and instruments used in the experiment should be sterile, and pay attention to the aseptic operation in the experiment.

2) Centrifugation temperature: some require room temperature; others require 4 degrees Celsius. The operation of the experiment should be as skilled and coherent as possible.

3) When separating mononuclear cells (PBMC) with Ficoll, it usually contains a small amount of red blood cells, which has little impact on the general experiment. If the experimental requirements are high, lysate can be used (some need to be sterile), and pay attention to the control of lysis time to avoid affecting the activity of mononuclear cells.

4) The effect of centrifugation with whole blood is worse than that of re centrifugation after whole blood dilution. The dilution is generally double the equal volume dilution.
