How to prepare high-quality serum samples by using separating gel to promote coagulation? Complete coagulation of blood and centrifugation conditions are two crucial links. Horizontal centrifuges are required for centrifugation.
The specific operation steps are as follows:
Immediately after blood collection, gently reverse the blood collection tube for 4~5 times to mix the samples. Wait for the samples to solidify fully. It needs to be placed for 30min, the centrifugation radius is 8cm, and the centrifugation speed is maintained at 3500~4000r/min for 10min. The serum and blood clot are completely separated by the separating gel, and the serum sample can be directly tested on the machine or transferred to the test cup matched with the instrument.
Only with this condition can high-quality serum samples be prepared, which shows that the separating gel has a good effect. If the centrifugation speed is too low, the force acting on the separation gel is relatively weak, the separation gel is not rotated well, or the blood is centrifuged without complete coagulation, fibrin condensates may stay in the serum or colloid layer, which may cause hemolysis. Except for emergency, the general biochemical examination has a good centrifugation effect after the blood is completely coagulated.
Due to lack of experience, this phenomenon often occurs in the initial use of separated gelled blood collection vessels in the laboratory. If fibrin filaments stay in the serum, it is easy to block the blood collection needle of the automatic analyzer. At present, the quality of many domestic separators has reached or approached the international level.
